A vapor barrier under a wooden floor or subfloor will trap the moisture that seeps through the concrete slab and will not allow the air to move. The moisture will build up. In addition, remember that the vapor barrier may deal with the moisture from the slab, but it is not addressing the moisture from other sources, which include plumbing leaks, hot water heater tanks, leaky washing machine hoses, etc. Should any of these accidents occur, your subfloor will be ruined. You will have to remove, discard, and replace it.
